Mouse Models for Investigating the Basis of Human Congenital
Cardiovascular Disease

Dr. Moon’s work has provided new insight into the pathways that regulate normal and abnormal development. By developing and employing advanced techniques for disrupting the function of genes in living mouse embryos, she generated a model system in which to study the roles of Fibroblast Growth Factors in diverse embryologic events, discovered critical roles for these genes during cardiac, pharyngeal and limb development, and provided new directions for investigating the molecular and genetic basis of human congenital syndromes, such as DiGeorge Syndrome.

Patient Visit to Research Career: Translational Work on Congenital Eye Movement Disorders

Dr. Elizabeth Engle is a child neurologist whose translational research has uncovered the genetic basis of a series of congenital ocular motility disorders and established that these disorders result from errors in cranial motor neuron development and axonal targeting within the brainstem and orbit. Her studies provide new avenues for the diagnosis and treatment of strabismus patients and for the study of neurodevelopmental disorders.

Gene Therapy for Inherited Lung Diseases

Dr. Flotte has developed the recombinant adeno-associated virus (rAAV) vector system for gene therapy of a number of chronic diseases affecting children, including cystic fibrosis (CF), alpha-1 antitrypsin (AAT) deficiency, type 1 diabetes, and metabolic disorders.  This work has spanned studies of the basic biology of AAV persistence and integration, the therapeutic effects of gene replacement in animal models of disease, preclinical safety testing of candidate vectors, and the first human trials of rAAV gene therapy in patients with CF and AAT efficiency.
